Assign audio suddenly not working

Hi,
I have been setting up sequences and assigning audio files OK for a few weeks now but suddenly it doesn't work. Even sequences that I had with an audio file assigned don't have the audio any more.
I know I am selecting the correct file because I have saved a new file with a different file name (using Audacity).
Any ideas gratefully received.

A bit more info -
If I open a sequence that previously worked fine with an audio file assigned and click on 'Visualiser' I get an error saying 'Object reference not set to an instance of an object'.
Assign audio shows nothing assigned.
